CD137 Signaling Is Critical in Fungal Clearance during Systemic Candida albicans Infection

چکیده

Invasive fungal infections by Candida albicans frequently cause mortality in immunocompromised patients. Neutrophils are particularly important for clearance during systemic C. albican infection, yet little has been known regarding which surface receptor controls neutrophils’ antifungal activities. CD137, is encoded Tnfrsf9, belongs to the tumor necrosis superfamily and shown regulate neutrophils Gram-positive bacterial infection. Here, we used genetic immunological tools probe involvement of neutrophil CD137 signaling innate defense mechanisms against We first found that Tnfrsf9−/− mice were susceptible whereas injection anti-CD137 agonistic antibody protected host from suggesting indispensable immunity Priming isolated with promoted their phagocytic fungicidal activities through phospholipase In addition, significantly augmented restriction growth received wild-type (WT) neutrophils. conclusion, our results demonstrate contributes infection promoting rapid clearance.

Host-Imposed Copper Poisoning Impacts Fungal Micronutrient Acquisition during Systemic Candida albicans Infections

Nutritional immunity is a process whereby an infected host manipulates essential micronutrients to defend against an invading pathogen. We reveal a dynamic aspect of nutritional immunity during infection that involves copper assimilation. Using a combination of laser ablation inductively coupled mass spectrometry (LA-ICP MS) and metal mapping, immunohistochemistry, and gene expression profiling...

Processing of Candida albicans Ece1p Is Critical for Candidalysin Maturation and Fungal Virulence

Candida albicans is an opportunistic fungal pathogen responsible for superficial and life-threatening infections in humans. During mucosal infection, C. albicans undergoes a morphological transition from yeast to invasive filamentous hyphae that secrete candidalysin, a 31-amino-acid peptide toxin required for virulence. Candidalysin damages epithelial cell plasma membranes and stimulates the ac...

Anti fungal activities of fresh garlic extract on candida albicans

Introduction: Denture stomatitis is one of the most problem in denture wearers and related to candida albicans. In This study we aimed to determaine garlic extract as an antifungal agent in oral candidiasis. Methods: Candida albicans growth in CHROM Agar was made serial dilution from 16 to 512 mg 1-1 from fresh garlic extract and measured minimmum Inhibitory Concentration. Normal salin w...

EGFR and HER2 receptor kinase signaling mediate epithelial cell invasion by Candida albicans during oropharyngeal infection.

The fungus Candida albicans is the major cause of oropharyngeal candidiasis (OPC). A key feature of this disease is fungal invasion of oral epithelial cells, a process that can occur by active penetration and fungal-induced endocytosis. Two invasins, Als3 and Ssa1, induce epithelial cell endocytosis of C. albicans, in part by binding to E-cadherin. However, inhibition of E-cadherin function onl...

Protection Against Epithelial Damage During Candida albicans Infection Is Mediated by PI3K/Akt and Mammalian Target of Rapamycin Signaling

BACKGROUND The ability of epithelial cells (ECs) to discriminate between commensal and pathogenic microbes is essential for healthy living. Key to these interactions are mucosal epithelial responses to pathogen-induced damage. METHODS Using reconstituted oral epithelium, we assessed epithelial gene transcriptional responses to Candida albicans infection by microarray. Signal pathway activatio...
